RESUMO

The aim of this work was to investigate the effects of dietary vitamin K3 (VK3) supplementation on production performance, egg quality, vitamin K-dependent proteins, and antioxidant properties in breeding geese during the laying period. A total of one hundred twenty 82-wk-old Wulong geese with similar body weights were randomly divided into 6 groups with 4 replicates and 5 geese each (1 male and 4 female). The geese in the control group were fed a basal diet, and the geese in the treatment groups were fed diets supplemented with different levels of VK3 (2.5, 5.0, 7.5, 10.0, and 12.5 mg/kg) for 11 wk. Dietary VK3 supplementation linearly and quadratically increased feed intake, egg mass, egg weight, and egg production (P < 0.05). Increasing VK3 levels linearly and quadratically increased albumen height, shell thickness and Haugh unit of eggs (P < 0.05). VK3 reduced osteocalcin (OC) and uncarboxylated osteocalcin (ucOC) levels in the serum. Dietary VK3 addition linearly decreased serum malondialdehyde (MDA) levels (P < 0.01). There was linear and quadratic effect in the activity of serum total superoxide dismutase (T-SOD) (P < 0.01), and linear effect in serum total antioxidant capacity (T-AOC) (P < 0.01). In conclusion, dietary VK3 supplementation enhanced the production performance, egg quality, vitamin K-dependent proteins, and antioxidant properties in breeding geese during the laying period. The optimal dose of dietary VK3 supplementation was 10.0 mg/kg.

This study investigated the effects of dietary arginine supplementation on the production performance, serum biochemicals, antioxidant capacity, and immunity of laying Wulong geese. A total of 150 Wulong geese (34-wk old) with similar body weights were randomly divided into 6 groups with 5 replicates and 5 geese each (1 male and 4 female). The geese in the control group were fed a corn-rapeseed meal basal diet, and the geese in the treatment groups were fed the basal diet supplemented with 0.1, 0.2, 0.3, 0.4, and 0.5% arginine. The experiment lasted for 17 wk. Our results showed that dietary arginine increased the egg production rate (LR) and average egg weight (AEW) of geese in a quadratic manner (P < 0.05). Dietary arginine had a quadratic effect on the contents of total protein (TP) and triglyceride (TG) (P < 0.05) in the serum. Dietary arginine quadratically decreased the content of malondialdehyde (MDA) and increased the activity of total superoxide dismutase (T-SOD) (P < 0.05). Dietary arginine supplementation linearly and quadratically increased the contents of immunoglobulin A (IgA) and immunoglobulin G (IgG), and linearly increased the content of nitric oxide (NO) (P < 0.05). In conclusion, dietary arginine supplementation can significantly improve the production performance, serum biochemicals, antioxidant capacity, and immunity of laying Wulong geese. Therefore, 0.3% arginine (actual content: 1.02%) is recommended in the diet.

The effects of unheated, 80 °C, 100 °C, and 121 °C on the physicochemical and microstructural properties of myofibrillary protein (MP) supplemented with glucose (Glc) and cellulose nanofibers (CNFs) were studied. The results showed that Glc and CNFs cross-linked with MP through non-covalent bonds when unheated compared with the unheated MP group, which increased the particle size and surface hydrophobicity of MP, and changed the secondary structure. At 80 °C and 100 °C, the particle size and hydrophobicity of MP significantly increased. At 121 °C, the contents of leucine and lysine in MP decreased by 16.2% and 19.9%, respectively, however, both Glc and CNFs could prevent this decrease. Meanwhile, microscopic images showed that the dispersion of Glc-MP and CNFs-MP were more uniform than the MP groups. Compared with Glc-MP, CNFs-MP had more uniform particle size distribution and better thermal stability at high temperatures (121 °C).

The effects of zinc (Zn) and Bacillus subtilis (B. subtilis) on reproductive performance, egg quality, nutrient digestion, intestine morphology, and antioxidant capacity were explored in geese breeders. Geese breeders (n = 120, 46-wk of age) were randomly assigned into 6 groups with 4 replicates of 5 birds each (1 male and 4 female). Breeders were fed diets with 2 levels of B. subtilis (2.5 × 109 and 5 × 109 CFU/kg) crossed with three levels of Zn (25, 45, and 65 mg/kg) for duration of 10-wk. The results showed that the egg laying rate (P < 0.05), fertility rate (P < 0.01), hatchability rate (P < 0.05), yolk color (P < 0.05), and the retentions of crude protein (P < 0.05), ether extract (P < 0.05) and phosphorus of geese breeders were improved by dietary supplementation of 5 × 109 CFU/kg B. subtilis and 25 mg or 45 mg/kg Zn. The serum T-SOD (P < 0.05) was increased by 45 mg/kg Zn supplementation. The serum T-AOC (P < 0.05) and retention of Zn (P < 0.05) were increased by 5 × 109 CFU/kg B. subtilis supplementation. The birds fed with 5 × 109 CFU/kg B. subtilis and 25 mg or 45 mg/kg Zn showed improved villus length (P < 0.01) and villus length/ crypt depth (P < 0.01) in both the jejunum and ileum. In conclusion, the combination of B. subtilis and Zn may have synergistic effects on these parameters, and dietary inclusion of 5 × 109 CFU/kg B. subtilis and 45 mg/kg Zn is recommended for improving the reproductive performance of geese breeders.

Vitamin D (VD) has the function of antibacteria and protect intestinal mucosa. Diacylglycerol has the property of dissolving VD, anti-bacterial, and antioxidant effects. The purpose of this study was to explore the potentially synergistic effects of diacylglycerol and VD in ameliorating dextran sodium sulfate-induced colitis in rats. A 2 × 3 factorial design was used in this experiment, consisting of two levels of VD (2.5 and 5 µg/day) crossed with three levels of duck oil diacylglycerol (0.5, 1, and 2 mL/day). The experiment lasted for 2 weeks. Compared with the colitis group, the physiological indexes were altered in colitis rats treated with diacylglycerol and VD, the concentrations of the pro-inflammatory indices were significantly reduced, the antioxidant enzyme activities were significantly increased, the diversity of caecal microflora was significantly increased. Besides, the expression of PPARγ was up-regulated while the expression of NF-κBp65 was downregulated. The changes of all those measures were toward those in the Healthy Control, and the mostly appropriate combination was 1 mL/day of diacylglycerol plus 2.5 µg/day VD.

This study investigated the effects of dietary supplementation of Bacillus subtilis-zinc on growth rates of the body and organs, nutrient utilization, microbial diversity in caecum, and contents of zinc (Zn) in organs of rats fed a Zn-deficient diet. In trial 1, 72 female pregnant SD rats were allocated to two groups and respectively fed the basal diet containing 13 mg Zn/kg, or the control diet containing 38 mg Zn/kg by supplementing ZnSO4 from day 10 of pregnancy until the offspring rats 24 days old. In trial 2, 18 offspring rats from the control group were fed the control diet. Ninety offspring from the Zn deficiency group were allocated to 5 groups and were fed the basal diet drenched with ZnSO4 solution (the total Zn 38 mg/kg), the basal diet only, or the basal diet drenched with Bacillus subtilis-Zn at doses of 2, 17, and 32 mg/kg Zn respectively. Each group had 3 replicates of 6 rats. The experiment lasted for 5 weeks. We found feeding young rats the Zn-deficient diet reduced feed intake, growth rate, unitization efficiencies of nitrogen (N), and Zn content in the organs. Administration of Bacillus subtilis-Zn to rats increased feed intake and the growth rates of the body and liver, kidney and heart, increased N utilization efficiency, and the contents of Zn in heart and brain but not in liver, regulated microbial diversity in the cecal content. The optimal amount of Bacillus subtilis-Zn ranged 15~20 mg Zn/kg, with the corresponding total Zn level at 28~33 mg/kg diet, the effects of which were superior to these by adding ZnSO4 at 38 mg/kg.

A total of 108 pregnant and lactation SD rats were divided into six groups, daily and orally dosed with Bacillus subtilis-copper (CuBs) and CuSO4 (CuS) both at three doses equivalent to 1.5 mg (Cu deficiency), 3 mg, and 6 mg Cu per kg diet, and the effects of the Cu source and dosage on the growth rate, immune status, duodenal development, and cecal microbial diversity were examined on 24-day-old offspring rats. The six offspring rats from each group were randomly selected for measuring the body weight gain and taking blood samples, and three rats were sacrificed for taking duodenum and cecum content samples. We found CuBs increased the body weight gain, development of duodenal villi, and survival rate of the offspring; increased the IgM content and lysozyme activity in serum; reduced the intestinal permeability; and increased the abundances of Lachnospiraceae, Ruminococcaceae, and Intestinibacter in the cecal content, when compared with CuS. We also found that Cu deficiency showed detrimental effects on the body weight gain and length, the survival rate of the offspring, and the immune indices in serum, as well as the increased intestinal permeability. We concluded that CuBs is better Cu source than CuSO4 for reproductive rats.

Iron deficiency anemia (IDA) is the most common nutritional deficiency worldwide. This deficiency could be solved by preparing stable, edible, and absorbable iron food ingredients using environmentally friendly methods. This study investigated enzymatic hydrolysis and microencapsulation process of goose blood. The physicochemical properties, stabilities of the microencapsulated goose blood hydrolysate (MGBH) and a supplement for rats with IDA were also evaluated. The results showed that the synergetic hydrolytic action of neutrase and alkaline protease significantly increased the heme-releasing efficiency. The heme was then microencapsulated using sodium caseinate, maltodextrin and carboxymethyl cellulose (CMC) as the edible wall material, and the encapsulation efficiency of the product reached 98.64%. Meanwhile, favorable thermal, storage and light stabilities were observed for the microencapsulation. It was found that MGBH can significantly improve the body weight and hematological parameters of IDA Wistar rat.

This experiment was conducted to study the effects of pantothenic acid on the growth performance, slaughter performance, lipid metabolism, and antioxidant function of one-to four-week-old Wulong geese and determine the appropriate supplemental level of pantothenic acid. A total of 360 one-day-old Wulong geese were randomly divided into 6 groups with 6 replicates per group and 10 geese (5 males and 5 females) per replicate. The geese in group Ⅰ (control group) were fed a basal diet, and the geese in groups Ⅱ to Ⅵ (experimental groups) were given the basal diet supplemented with 8, 15, 30, 60, and 120 mg/kg pantothenic acid, respectively. The experiment lasted for 4 weeks. Based on the results of unrelated comparative analysis, the body weight was the highest when the dietary pantothenic acid level was 27.57 mg/kg. When the dietary pantothenic acid level was 26.17 mg/kg, the average daily gain peaked. When the dietary pantothenic acid level was 15.50 mg/kg, the feed:gain ratio was the lowest. The percentage of abdominal fat in groups Ⅲ and Ⅳ was significantly lower than that in group Ⅰ (P < 0.05). The content of total cholesterol in serum in groups Ⅲ to Ⅴ was significantly lower than that in group Ⅰ (P < 0.05). The triglyceride content in groups Ⅲ and Ⅳ was significantly lower than that in group Ⅰ (P < 0.01). The high-density lipoprotein-cholesterol content in group Ⅳ was significantly higher than that in group Ⅰ (P < 0.05). The total antioxidant capacity of serum and liver in group Ⅳ was significantly higher than that in group Ⅰ (P < 0.05). The malondialdehyde content in the liver in groups Ⅲ and Ⅳ was significantly lower than that in group Ⅰ (P < 0.05). Glutathione peroxidase activity in the serum in group Ⅳ was significantly higher than that in group Ⅰ (P < 0.05). Glutathione peroxidase activity in the liver in groups Ⅳ and Ⅴ was significantly higher than that in group Ⅰ (P < 0.01). The addition of pantothenic acid in the diet of one-to four-week-old Wulong geese significantly affected the growth performance, slaughter performance, lipid metabolism, and antioxidant function of the geese. In terms of economic benefits, the optimal supplemental level of pantothenic acid in one-to four-week-old geese was 15.50 mg/kg.

Major histocompatibility complex (MHC) was correlated with immune response for extra-antigen. MHC sequences of chicken, other birds, crawl species and mammalian were derived from GenBank/DDBJ/EMBL and analyzed by alignment, and then primers were designed. By means of LA-PCR method, MHC Classgene was cloned from Wulong goose genomic DNA and total RNA, and the structure of MHC Classgenomic DNA was analyzed using bioinformation methods. The results showed that the genomic DNA, which consisted of 8 extrons and 7 introns, had 64.1% amino acids homologous to chicken and 42.9% amino acids to human. The molecular phylogenetic tree further revealed evolutionary relationship between Wulong goose and other animals such as chicken, other birds, crawl species, mammalian and human. The results of homologous modeling showed that Wulong Goose MHC Classgene was formed by amidocyanogen (N) terminal structure region and carboxyl (C) terminal structure region.
